Humoral Immune Response against a Hepatitis C Virus Envelope E2 Variant Expressed in Escherichia coli

چکیده
A hepatitis C virus (HCV) E2 variant encompassing amino acids 458 to 680 was expressed in Escherichia coli. The recombinant product (His-E2680) was immunoidentified as a 30-kD protein using the serum from a patient with chronic hepatitis C, and was purified using Ni-NTA chelate chromatography. When HCV-positive human sera were tested with the His-E2680 protein, 39% of them showed anti-E2 reactivity. ELISA experiments showed that this antigen was very immunogenic in immunized rabbits. Using synthetic peptides, we identified an immunologically relevant region in the 643–671 segment of the His-E2680 protein.

Cloning and Purification of the Hydrophilic Fragment of Hepatitis C Virus E2 Protein Fused to the Choline-binding Domain of the Major Autolysin of Streptococcus pneumoniae: Evaluation of the Humoral Immune Response in Rabbits
The hydrophilic fragment of hepatitis C virus (HCV) E2 protein was expressed in Escherichia coli as a chimera, bound to the choline-binding domain (C-Lyt) of the major autolysin of Streptococcus pneumoniae. The C-LytE2 chimera was purified by affinity chromatography using DEAE-Sepharose. The purified antigen was used to immunize rabbits and the specific humoral immune response to E2 protein was...
متن کاملEvaluation of full length E1 and E2 glycoproteins of HCV expressed in P. pastoris as a protein-based vaccine candidate
Introduction: The development of an effective vaccine against Hepatitis C virus (HCV) is still a target of intensive vaccine research. The HCV envelope proteins E1 and E2 which can induce broadly neutralizing antibodies are the major candidate for this purpose. Different types of expression systems have been used to express these glycoproteins. In this study, an expression system using Pichia p...
متن کاملIncomplete humoral immunity against hepatitis C virus is linked with distinct recognition of putative multiple receptors by E2 envelope glycoprotein.
Little is known about the role of the humoral immune response to hepatitis C virus (HCV). This study provides molecular evidence for the mechanism by which neutralizing Abs from the sera of chronic HCV patients have lower inhibitory activities against the binding of HCV E2 envelope protein to human hepatoma cell lines than to a lymphoma cell line. E2 binds to several putative receptors, specifi...
متن کاملEffect of IL-2 co-expressed or co-inoculated with immuno-dominant epitopes from VP1 protein of FMD virus on immune responses in BALB/c mice
Objective(s): The results of studies on vaccine development for foot-and-mouth disease (FMD) virus show that the use of inactivated vaccines for FMD virus is not completely effective. Novel vaccinations based on immuno-dominant epitopes have been shown to induce immune responses. Furthermore, for safety of immunization, access to efficient adjuvants against FMD virus seems to be critical.Materi...
متن کاملDevelopment of Preventive Vaccines for Hepatitis C Virus E1/E2 Protein
Hepatitis C virus (HCV) is responsible for a vast majority of liver failure cases. HCV is a kind of blood disease appraised to chronically infect 3% of the worlds’ population causing significant morbidity and mortality. Therefore, a complete knowledge of humoral responses against HCV, resulting antibodies, and virus-receptor and virus-antibody interactions, are essential to design a vaccine. HC...
